Stage 4 traced a route from Norwood to Tanunda over 130km.
André Greipel lost more than 7 minutes by the end of the stage, surrendering the ochre jersey to Martin Kohler of Team BMC.
Rohan Dennis of Team Uni SA snuck off the front to take the KOM points on offer on Mengler Hill.
Óscar Freire of Team Katusha turned on the afterburners to take the stage win in Tanunda.
